Unhappy Answered: Asking about Deuterium NMR

I wanted to do Deuterium NMR in human urine sample but no one has done previously in our lab.
In our Lab. we have 400 MHz Bruker (Avance) spectrometer with BroadBand Inverse probe (1H Observe, Z-gradient).
How we have to proceed without deuterium lock which is neccessory for exp. ?
Is above described instruments specification sufficient to perform the deutrium NMR experiments ?

Please any on can help

Thanks very much

Do you have your human urine sample labeled with deuterium !!! Just kidding.... All you have to do is switch off the lock using command "locnuc=off" and use the sequence ZG2H (read the sequence carefully). Well, this is a general instruction to run 2H spectrum in a Bruker spectrometer. If you fry the probe without taking proper care, i am not responsible for it !!!

Smile Thanks

Thanks Man,
Is it require proton decoupling ?
if yes.
then tell me that zgdc or zgpg pulse sequence can be used or not ?

An alternative method to switching off the D-Lock is the use of capilar tubes, I use a NewEra's capilar tube for 500 µL of sample and 60 µL of D2O in the capilar

[Question from NMRWiki Q&A forum] Should I decouple deuterium on 2H/15N labeled sample for R1, R2 and hetnoe?
Should I decouple deuterium on 2H/15N labeled sample for R1, R2 and hetnoe? If I'm running relaxation experiments (15N) for R1, R2, and hetnoe on a double labeled (2H/15N) sample, do I need to do any deuterium decoupling added to the program? Check if somebody has answered this question on NMRWiki QA forum

[Question from NMRWiki Q&A forum] What has been done with direct detection of Deuterium?
What has been done with direct detection of Deuterium? Hello, I am trying to investigate what has been done in NMR using direct detection of deuterium in liquids. - Could you suggest any citations? What kind of sensitivity gain there is in using broadband probe vs. lock channel? Has anything been done/can be done to detect scalar couplings with protons like in COSY or TOCSY? Are HD couplings other than geminal detectable? Thanks.

[U. of Ottawa NMR Facility Blog] Deuterium Decoupling
Deuterium Decoupling Bruker NMR spectrometers equipped with a 2H TX board and 2H amplifier can be setup to carry out 2H observation and decoupling experiments while still maintaining a 2H lock. Two channel systems can use the 2H transmitter for 2H, 1H, 2H and X measurements. In addition to these, a three channel system can be set up for X measurements. The first figure below shows the 13C and 13C spectra collected for a mixture of deuterated solvents using two channels of an AVANCE spectrometer. The second figure shows the 13C, 13C, 13C and 13C spectra for the standard 13C sensitivity...
