Default Internal pH indicators for biomolecular NMR

Internal pH indicators for biomolecular NMR

Olga K. Baryshnikova, Thomas C. Williams and Brian D. Sykes
Journal of Biomolecular NMR; 2008; 41(1); pp 5-7

Abstract:
We describe a non-invasive technique for determining pH in biomolecular NMR sample using buffer components (formate, tris, piperazine, and imidazole) as internal pH indicators, whose 1H NMR chemical shifts are sensitive to pH in a range from 2.5 to 9.8. This method is suitable for a wide range of applications where samples are handled intensively during NMR titrations or in high throughput analysis in structural genomics or metabolomics.

J Am Chem Soc. 2002 May 22;124(20):5881-9 Authors: Schleucher J, Wijmenga SS NOESY and ROESY cross-peak intensities depend on internuclear distances and internal motion. Internal motion is usually ignored, and NOESY cross-peak intensities are interpreted in terms of internuclear distances only. Off-resonance ROESY experiments measure a weighted average of NOE and ROE. The weight can be described and experimentally set by an...

[Question from NMRWiki Q&A forum] Internal referencing in direct and indirect dimensions using guanidinium
Internal referencing in direct and indirect dimensions using guanidinium Hello, I am recording 15N-HSQC spectra of a protein in increasing concentrations (0.25–2.5 M) of guanidine hydrochloride. I would like to use the guanidine chemical shift (which appears in the HSQC) as an internal standard in both the direct and indirect (!) dimensions. My intuition is that the chemical shifts of guanidine should be independent of guanidine concentration. Does anyone know if this is really true? Currently, I see a constant upfield shift in the indirect dimension, even for the guanidine peak...
